Previous studies have shown increased glucose uptake in vascular smooth muscle (VSM) cells of spontaneously diabetic rats (SDR) in vivo. In this study, we examined whether glucose uptake in VSM cells and platelets of SDR was altered in vitro. Glucose uptake by VSM cells was determined using 2-[3H]deoxy-D-glucose. Glucose uptake by platelets was measured by [3H]D-glucose binding. No significant difference was observed in the basal uptake of [3H]2-deoxy-D-glucose by VSM cells or platelets of SDR and control rats. Incubation of VSM cells or platelets with 30 mM or 5 mM glucose resulted in a significant increase in 2-[3H]deoxy-D-glucose uptake in both SDR and control rats. Addition of insulin to cells or platelets at a concentration of 100 nM resulted in a significant decrease in 2-[3H]deoxy-D-glucose uptake in both SDR and control rats. Although insulin increased glucose uptake in VSM cells of control rats, the effect was not observed in SDR. The results indicate that glucose uptake by VSM cells and platelets is increased in SDR in vitro. This increase is related to insulin resistance.Q:
